PEPN1924 - A Phase 1/2 study of DS-8201a (NSC# 807708, IND# 153036) in children, adolescents, or young adults with recurrent Osteosarcoma, Wilms Tumor, and Desmoplastic Small Round Cell Tumor
This phase II trial studies the effects of trastuzumab deruxtecan in treating patients (> 12 years and ≤ 39 years of age) with HER2 positive osteosarcoma that is newly diagnosed or has come back (recurrent). Trastuzumab deruxtecan is a monoclonal antibody, called trastuzumab, linked to a chemotherapy drug called deruxtecan. Trastuzumab attaches to HER2 positive cancer cells in a targeted way and delivers deruxtecan to kill them.
• children, adolescents, and young adults with Wilms tumor or desmoplastic small round cell tumor (DSRCT)
• disease that has returned after treatment or has not responded to standard treatment
• measurable tumor that can be evaluated during the study
• recovered from previous cancer treatment and meet study health requirements

A Phase I/IIa, First-in-Human, Multicenter, Monotherapy and Combination-Therapy with Nivolumab, Dose-Finding and Dose-Expansion Study of (212Pb)VMT01 Melanocortin-1-Receptor-Targeted, Image-Guided, Alpha-Particle Therapy in Subjects with Previously Treated Unresectable or Metastatic Melanoma (VMT01-T101)
This study is testing a radioactive treatment for adults with advanced melanoma that has spread, cannot be removed by surgery, or has gotten worse after prior treatment. We want to learn how safe the treatment is and how well it may work. By taking part, participants may help us learn whether this could become a useful treatment option for people with advanced melanoma.
• ages 18 or older
• diagnosed with advanced melanoma that cannot be removed by surgery, has spread, or has returned
• previously received at least one approved treatment for advanced melanoma and had the cancer worsen

Targeted Oligometastatic Radiation in Pediatric and Young Adult Patients with Soft Tissue and Bone Sarcoma (TARGET-RT) (TARGET-RT)
This study aims to improve treatment and outcomes for children, adolescents, and young adults with metastatic sarcoma and help guide future care for this disease.
• younger than 39 years old or were first diagnosed with a soft tissue or bone sarcoma before age 39
• have a newly diagnosed, recurrent, or progressive (continued to grow) soft tissue or bone sarcoma
• have sarcoma that has spread (metastasized) to another part of the body and can be measured on imaging scans

• have metastatic disease that cannot be measured on imaging scans

COG ARST2032: A Prospective Phase 3 Study of Patients with Newly Diagnosed Very Low-risk and Low-risk Fusion Negative Rhabdomyosarcoma
Rhabdomyosarcoma is a type of cancer that occurs in the soft tissues in the body. This phase III trial aims to maintain excellent outcomes in patients with very low risk rhabdomyosarcoma (VLR-RMS) while decreasing the burden of therapy using treatment with 24 weeks of vincristine and dactinomycin (VA) and examines the use of centralized molecular risk stratification in the treatment of rhabdomyosarcoma. Another aim of the study it to find out how well patients with low risk rhabdomyosarcoma (LR-RMS) respond to standard chemotherapy when patients with VLR-RMS and patients who have rhabdomyosarcoma with DNA mutations get separate treatment. Finally, this study examines the effect of therapy intensification in patients who have RMS cancer with DNA mutations to see if their outcomes can be improved.
• 21 or younger at time of enrollment
• newly diagnosed embryonal rhabdomyosarcoma (ERMS), spindle cell/sclerosing RMS, or FOXO1 fusion negative alveolar rhabdomyosarcoma
• must be enrolled in APEC14B1 (NCT02402244) prior to enrollment and treatment on ARST2032 (this trial)
• contact study team for more detailed criteria
• received prior chemotherapy and/or radiation therapy for cancer prior to enrollment
• unable to undergo radiation therapy
• Females who are pregnant
